          【】 目的 探讨血浆凝血酶激活的纤溶抑制物(TAFI)在2型糖尿病大鼠肾损伤发病机制中的作用。方法 选用一月龄成年雄性 Wistar 大鼠36只,每组12只。并随机分成三组:正常组(A)、糖尿病假手术组(B)、糖尿病手术组(C)。A组予以普通饲料喂养并腹腔注射柠檬酸缓冲液,B、C组予以高糖高脂饲料喂养及腹腔注射链脲佐菌素(STZ),其中B组行假手术,C组予以右肾切除术诱导2型糖尿病肾病模型.每组检测相关指标并取肾脏组织,作HE病理组织染色,观察肾脏病变情况。结果 C组的体重、肾指数、血糖(FBG)、胰岛素(INS)、血肌酐(SCr)、血脂、24 h尿蛋白、TAFI等水平明显高于A、B组,病理显示明显的肾脏病变。结论 三联法(高糖高脂饮食+单侧肾脏切除+小剂量链脉佐菌素STZ)能建立稳定的符合2型糖尿病肾病大鼠模型;TAFI参与2型糖尿病大鼠肾损伤发生发展。

          【】 Objective To explore the effect of Plasma thrombin activated Fibrinolysis inhibitor (TAFI) in the pathogenesis of type 2 diabetic rats kidney injury. Methods Select 36 adult male Wistar rats who is a-month old,12 in each group, and randomly divided into three groups: normal diet group(A)、sham operation group (B)、operation group (C). Feed group A with common fodder and inject critric acid buffer by enterocoelia. Feed group B and C with high fat and sugar diet and then inject STZ by enterocoelia. Among them, give group B sham operation, but give group C surgical removal of the right kidney to induct the model of type 2 diabetic nephropathy. Test three group’s relevant indexes and take nephridial tissue to do HE pathological staining and observe it. Result Weight, kidney index, blood glucose (FBG), insulin (INS), serum creatinine (SCr), blood lipid, 24h urine protein, TAFI level in group C were obviously higher than that of group A and B, pathology showed obvious kidney disease. Conduction triplex process (high fat and sugar diet unilateral nephrectomy a low dose of STZ ) can build out of the steady rat model which accords with the characteristics of type 2 diabetic nephropathy; TAFI have participated in the development of the kidney injury of type 2 diabetic rats .
