          目的 探讨PI3K/Akt/mTOR通路对大鼠肾小球系膜细胞增殖的调控作用。方法 体外培养大鼠系膜细胞,随机分为正常对照组、表皮生长因子(EGF)(10ng/mL)组、LY294002[2ug/ml]组、EGF联合LY294002组,采用四甲基偶氮唑蓝(MTT)法检测各组系膜细胞的增殖,流式细胞术检测各组系膜细胞周期,免疫荧光法检测各组系膜细胞mTOR的表达。结果 与正常对照组相比,EGF组能显著促进大鼠系膜细胞增殖,G0/G1期细胞减少,S期及G2/M期细胞增加,且mTOR表达增加,LY294002组大鼠系膜细胞增殖受抑制,G0/G1期细胞增加,S期及G2/M期细胞减少,且mTOR表达降低;与EGF组比较,EGF联合LY294002干预组系膜细胞增殖受抑制,G0/G1期细胞增多,S期及G2/M期细胞减少,且mTOR表达降低。结论 mTOR信号通路参与了大鼠肾小球系膜细胞增殖的调控,抑制该信号通路的活化可显著抑制系膜细胞的增殖,并抑制系膜细胞周期的进程。

          Objective To investigate the role of PI3K/Akt/mTOR signaling pathway in the process of regulating rat glomerular mesangial cell proliferation. Method The cultured rat glomerular mesangial cells were randomly divided into four groups: normal control group, epidermal growth factor (EGF) (10ng/ml) group, LY294002 (2ug/ml, the inhibitor of PI3K) group and EGF combine LY294002 group. They were treated for 48 hours. The cells proliferation was measured by MTT colorimetric method. The cell cycles were measured by flow cytometry. The expression of mTOR in each group was measured by immunofluorescence staining. Results Compared with the normal control group, the EGF group can promote the proliferation of rat glomerular mesangial cells. The cells of G0/G1 phase were decreased and the cells of S and G2/M phase were increased. The expression of mTOR was increased. The LY294002 group can inhibit the proliferation of rat glomerular mesangial cells. The cells of G0/G1 phase were increased and the cells of S and G2/M phase were decreased. The expression of mTOR was decreased. Compared with the EGF group, the EGF combine LY294002 group can inhibit the proliferation of rat glomerular mesangial cells. The cells of G0/G1 phase were increased and the cells of S and G2/M phase were decreased. The expression of mTOR was decreased. Conclusion mTOR signaling pathway take part in the process of rat glomerular mesangial cell proliferation, inhibiting the signaling pathway can significantly inhibit the proliferation and cell cycle of rat glomerular mesangial cells.
