          目的 探讨RSVA314对白血病细胞K562 细胞迁移和增殖的影响,为RSVA314可能治疗白血病提供实验依据。方法 通过体外培养K562 细胞,实验分为四组:生理盐水对照组、RSVA314低剂量组(1 μM)、RSVA314中剂量组(5 μM)、RSVA314高剂量组(25 μM)。采用CCK-8方法检测K562细胞的增殖活性;采用Transwell 迁移实验检测K562细胞的迁移能力;采用Western blot检测K562细胞蛋白激酶B(AKT)的表达。结果 与对照组相比,RSVA314中、高剂量组OD值明显增加(P<0.05,P<0.01),提示其可抑制K562细胞的增殖活性。RSVA314低、中、高剂量组穿膜细胞数量明显减少,与对照组相比有明显的差异(P<0.05,P<0.01,P<0.01)。Western blot 检测显示RSVA314中、高浓度组可抑制AKT磷酸化的表达,与对照组相比有显著差异(P<0.05,P<0.01)。

          ABSTRACT: Objective To explore the effect of RSVA314 on the migration and proliferation of leukemic K562 cells and to provide experimental data for the treatment of leukemia with RSV314. Methods The leukemic K562 cells were cultured in vitro and there were four groups in this experiment: Control group, low dose of RSVA314(1 μM); middle dose of RSV314 group(5 μM)and high dose of RSVA314 group(25 μM).The proliferation activity of leukemic K562 cells was detected by measuring the OD value with the spectrophotometer. Transwell migration assay was used to detect the migration ability of K562 cells. The expression of AKT in K562 cells was detected by Western blot. Results The OD value increased significantly in the middle and high dose of RSVA314 group (P < 0.05, P < 0.01, as compared to the control group), indicating that RSV314 can inhibit the proliferation of K562 cells. Compared with the control group, the ability of migration in each RSVA314 treated group was inhibited (P<0.05, P < 0.01). The results of Western blot showed that RSVA314 could inhibit the expression of p-AKT in the middle and high concentration group, and there was significant difference as compared with the control group (P<0.05, P<0.01). Conclusion RSVA314 can inhibit the migration and proliferation of leukemic K562 cells partly by inhibiting the expression of p-AKT.
