          目的: 研究白花蛇舌草对病毒肺炎小鼠免疫细胞因子表达的影响。方法:选取48只昆明种小鼠,分为正常组、模型组、白花蛇舌草组、病毒唑组,每组12只。模型组、白花蛇舌草组和病毒唑组均以25μL(流感病毒新鲜尿囊液10倍系列稀释成1×10-4 )流感病毒滴鼻感染小鼠,构建肺炎小鼠模型。构模后,白花蛇舌草组以3.0825g/kg白花蛇舌草灌胃;病毒唑组以10mg/kg病毒唑灌胃,均每天灌胃1次,连续7d。正常组小鼠以等量的生理盐水灌胃处理。7d后,称取体质量和肺组织质量,计算肺指数。收集血液和肺组织,用酶联免疫吸附法(ELISA)测定TNF-α、IL-6含量。收集肺泡灌洗液,计数白细胞数及分类。用Western blot方法测定肺组织中Toll样受体4(TLR4)、核因子-κBp65(NF-κB p65)、细胞间粘附分子-1(ICAM-1)、血管细胞粘附分子-1(VCAM-1)蛋白水平。结果: 与正常组比较,模型组小鼠肺指数升高,血和肺组织中TNF-α、IL-6含量升高,肺泡灌洗液中白细胞总数、单个核细胞数、中性粒细胞数目均增多,肺组织中TLR4、NF-κB p65、ICAM-1、VCAM-1蛋白表达水平升高(均P<0.05)。与模型组比较,白花蛇舌草组和病毒唑组小鼠肺指数降低,血清和肺组织中TNF-α、IL-6含量降低,肺泡灌洗液中白细胞总数、单个核细胞数、中性粒细胞数目均减少,肺组织中TLR4、NF-κB p65、ICAM-1、VCAM-1蛋白表达水平降低(均P<0.05)。结论: 白花蛇舌草可以降低肺炎小鼠免疫细胞因子表达,其机制可能与TLR4/NF-κB信号轴和粘附分子表达有关。

          Objective: To study the effect of Hedyotis diffusa willd on the expression of immune cytokines in mice with pneumonia. Methods: A mouse model of pneumonia was established by infecting mice with influenza virus nose drop infection of 25μL (diluted to 10-4), the stomach was treated with Hedyotis diffusa Willd (3.0825g/kg, 200μL, gavage) once a day for 7 days. The body weight and lung tissue mass of mice were weighed, the lung index calculated, and serum and lung tissue were collected. The contents of TNF-α and IL-6 were determined by ELISA. Bronchoalveolar lavage fluid was collected, and leukocytes were counted and classified. The levels of TLR4, NF-κB p65, ICAM-1 and VCAM-1 proteins in lung tissues were measured by Western blotting. Results: As compared with normal mice, the lung index of mice with influenza virus pneumonia, the levels of TNF-α and IL-6 in serum and lung tissues, the total number of leukocytes, the number of mononuclear cells and the number of neutrophils in the bronchoalveolar lavage fluid, and the expression levels of TLR4, NF-κ B p65, ICAM-1 and VCAM-1 protein in lung tissue increased. Treatment with Hedyotis diffusa willd could reduce the lung index of pneumonia mice, the levels of TNF-α and IL-6 in serum and lung tissue, the total number of leukocytes, the number of mononuclear cells and the number of neutrophils in bronchoalveolar lavage fluid, and the expression levels of TLR4, NF-κB p65, ICAM-1 and VCAM-1 in lung tissue. Conclusion: Hedyotis diffusa willd can reduce the expression of immune cytokines in pneumonia mice, which may be related to TLR4/NF-κ B signal axis and adhesion molecules.
